Hundreds of thousands of individuals have been sequenced, providing an exciting resource to improve the clinical diagnosis, novel therapeutics and precision medicine. I will describe how we built a knowledgebase through sequencing cohorts, literature mining, and manual curation, and developed various applications to interpret genomes with electronic medical records for diagnosis, driver gene discovery, and novel therapeutics on cardiovascular disease, Alzheimer's diseases, and endocrine tumors. I will also discuss how we create personalized cancer therapeutic reports for cancer patients at Mount Sinai hospital.
