Exploring the Chemodiversity and Antitumor Activities of Filamentous Fungi

The silent biosynthetic pathways of two Tolypocladium sp.fungal strains were activated by microbial co-culture and chemical epigeneticsapproaches resulting in the identification of novel secondary metabolites, maximiscin and gichigamin A, with unusual structural skeletons. By exploring the biosynthetic pathway of the unique shikimate-PKS-NRPS hybrid metabolite maximiscin, a novel microbial chemical defense mechanism was disclosed. The maximiscin-producing fungus utilizes the electrophilic natural products, pericoxide and pericosine A, as chemical warheads to neutralize disparate antibiotics and toxins. The first 22-mer peptaibol-class linear peptide, gichigamin A, showedunexpectedly high degree of cell membrane penetration and elevated in vitro tumor cell cytotoxicity. One-step modification of the C-terminus of gichigamin A generated a coumarin adduct with 100-fold-increased cytotoxicity. The coumarin adduct exhibited an altered mechanism of cytotoxicity that enhanced cytoplasmic delivery and eliminated the liability of ion leakage associated with natural peptaibols.

杜林,男,1982年出生,现为美国俄克拉荷马大学研究助理教授。2004年于中国海洋大学药学专业取得学士学位,2009年于中国海洋大学药物化学专业取得硕博连读博士学位。2011年至2014年期间分别于美国密西西比大学、美国俄克拉马荷大学从事博士后研究,2015年于美国俄克拉马荷大学任教至今。主要研究方向为天然产物化学、新药开发以及相关方向。主要获得荣誉有美国生药学会年会会员旅游奖(2013年),山东省优秀博士论文学位奖(2010年),中国海洋大学优秀博士论文学位奖(2009年),中国国家奖学金(2003)等等。研究成果发表学术论文40余篇,主要论文有Chemoreactive Natural Products Afford Resistance gainst Disparate Antibiotics and Toxins.
